Ple139-lacZ Description:

The Ple139 promoter construct is derived from the promoter region of the Orphan Nuclear Receptor NR2E1 (NR2E1) gene. The Pleiades Promoter Project created a 4743 bp MiniPromoter derived from four upstream putative regulatory subregion along with a section of the promoter and introduced it into the multiple cloning site (MCS) of Pleiades expression vector pEMS1313 (LacZ reporter flanked by full frt sites (WT and F5 mutant)) to make plasmid pEMS1611 with Ple139 driving lacZ. The Ple139-lacZ (pEMS1611) was introduced by homologous recombination as single copy insertion into the Hprt1b-m3 locus in the X Chromosome of mouse embryonic stem cells (ESCs) mEMS1202. The resulting PCR validated two ESC lines, mEMS4526, and mEMS4523. Chimeras were generated using mEMS4526.

Note: Promoterless negative controls (pEMS1302, pEMS1306, pEMS1313) have been generated, as well as CAG positive controls (pEMS1157, pEMS1277 & pEMS1488).
